James Abankwah (Ireland U21s), Michael Hector (Jamaica), Terell Thomas (Saint Lucia) and Karoy Anderson (Jamaica) have all linked up with their respective countries for their upcoming international matches.
Harry Isted (knee), Tayo Edun (hamstring), Conor McGrandles (knee), Terry Taylor (ankle) and Panutche Camará (foot) are also unavailable for Tuesday's cup clash.
The young Villans kicked off their EFL Trophy campaign with a 2-2 draw at Sutton United, before going on to win 5-4 on penalties to claim an extra point.
Tommi O'Reilly and Omari Kellyman gave Villa a two-goal lead on the night, but the visitors pegged them back with two late strikes - including Harry Beautyman's leveller 10 minutes into added time.
That result leaves Inigo Idiakez’s side sat second in Group B ahead of Tuesday's meeting in SE7.
The two sides have gone head-to-head on one previous occasion. That meeting came at The Valley in this same competition in November 2021, when goals from Mason Burstow and Jayden Stockley earned the Addicks a 2-1 triumph and secured their place in the last 16.
Neil Hair will control proceedings on Tuesday. The official last refereed a Charlton fixture in October 2022 - a home fixture against MK Dons.
Hair will be assisted by Paul Evans and Laurentiu Hantea, while Lisa Benn is the Fourth Official.
