Appleton is 'reasonably confident' Lloyd Jones, Tayo Edun and George Dobson will be available for the Addicks after the trio were withdrawn at the Select Car Leasing Stadium on Wednesday.
Harry Isted (knee), Conor McGrandles (calf), Terry Taylor (ankle), Panutche Camará (hamstring), Chuks Aneke (hamstring) and Miles Leaburn (hamstring) remain sidelined.
The visitors will check on the fitness of top scorer Fejiri Okenabirhie (quad), who was forced off after 22 minutes on Saturday, though Harrison Dunk (knee) is unavailable.
Neil Harris was named as Cambridge's new head coach on an 18-month contract on Wednesday. The U's currently sit 18th in the Sky Bet League One table, four points above the relegation zone.
Cambridge claimed an emphatic 4-0 triumph at home to Fleetwood Town in the second round of the Emirates FA Cup last weekend, with Danny Andrew, Elias Kachunga, Okenabirhie and Gassan Ahadme all on the scoresheet.
That result was preceded by a run of three successive league defeats against Peterborough United, Northampton Town and Lincoln City respectively.
Charlton and Cambridge have locked horns on 22 previous occasions with the Addicks winning eight, the U’s seven and the remaining seven meetings resulting in draws.
The two teams last clashed in March 2023, when first-half strikes from Leaburn and Jes Rak-Sakyi earned Charlton a 2-1 victory in Cambridgeshire.
Thomas Kirk will control proceedings on Saturday. The official has refereed 14 fixtures so far this season, showing 62 yellow cards and one red card.
Kirk will be assisted by Daniel Cook and Joseph Stokes, while Damith Bandara is the Fourth Official.
